@#【Objective】The aim of this study was to investigate the associations between serum complement C3,C4 and low density lipoprotein cholesterol(LDL- C)levels and early- onset coronary heart disease.【Methods】We enrolled 255 cases of coronary angiography confirmed coronary artery disease from January 2018 to September 2018. All the patients were divided into early- onset coronary heart disease group(108 cases)and late- onset coronary heart disease group(147 cases). Besides ,100 healthy subjects were enrolled and used as controls. Serum levels of C3 ,C4 and LDL-C were analyzed by automatic biochemical analyzer.【Results】Levels of serum C3,C4 and LDL-C in early-onset coronary heart disease group,late-onset coronary heart disease group and healthy control group were significantly different(P < 0.05). In early-onset coronary heart disease group,C3 and C4 were positively correlated with LDL-C(P < 0.05). However ,there was no significant correlation (P > 0.05) between C3 ,C4 and LDL- C in late- onset coronary heart disease group and healthy control group.【Conclusions】The levels of C3 and C4 were positively correlated with LDL-C only in the early-onset coronary heart disease patients.

ABSTRACT

El objetivo del presente trabajo fue la estimación del intervalo de referencia para los componentes del complemento C3 y C4 en población adulta hospitalaria. Se siguieron los lineamientos de la guía C28A3 de CLSI para lo cual se eligió como población de referencia a dadores de sangre concurrentes al Servicio de Hemoterapia del hospital que superaron el interrogatorio médico y accedieron a la extracción. Fue seleccionada una muestra de 251 dadores constituida por 72,9% de mujeres, de edad promedio 36,5±10 años y 27,1% de hombres, de edad promedio 40,6±11,5 años. Esta composición estuvo balanceada por sexo y edad promedio a la población de pacientes que asisten al laboratorio, en su mayoría con diagnóstico presuntivo de enfermedades autoinmunes. Los analitos fueron dosados por nefelometría cinética con nefelómetro Immage 800 de Beckman Coulter (California, EE.UU.). El intervalo de referencia se calculó por el método no paramétrico, es decir, se estimó el intervalo de confianza del 95% central de cada distribución de valores. Los límites obtenidos fueron: IC95% C3=70-165 mg/dL IC95%, C4=14-37 mg/dL. Estos resultados fueron posteriormente verificados con una serie de 20 nuevos dadores y fueron comparables a valores obtenidos en otras series citadas en la literatura.


The aim of this study was to estimate the reference interval for the components C3 and C4 complement in an adult population. The guidelines of the C28A3 document CLSI were followed, for which blood donors attending to a hospital blood centre who passed the medical examination and agreed to extraction were chosen as reference population. A sample constituted by 251 donors, 72.9% women, average age 36.5±10 years and 27.1% men, average age 40.6±11.5 years was selected. This composition was balanced by gender and average age to the population of patients attending the laboratory, mostly with autoimmune diseases. The analytes were measured by rate nephelometry with Immage 800 Nephelometer, Beckman Coulter (California, USA). The reference range for the non-parametric method was calculated, this is to say, the 95% central confidence interval of each value distribution was estimated. The limits obtained were: 95% CI C3=70-165 mg/dL and 95% CI C4=14-37 mg/dL. These results were later verified with a series of 20 new donors and are comparable to values obtained in other studies cited in the literature.


O objetivo deste estudo foi estimar o intervalo de referência para os componentes do complemento C3 e C4 na população adulta hospitalar. Foram seguidos os lineamentos do guia C28A3 de CLSI para o qual foi escolhida como população de referência doadores de sangue concorrentes do Serviço de Hemoterapia do hospital que passaram o questionário médico e acederam à extração. Foi selecionada uma amostra constituída por 251 doadores, 72,9% mulheres, com idade média de 36,5±10 anos e 27,1% dos homens de idade média 40,6±11,5 anos. Esta composição esteve equilibrada por sexo e idade média de pacientes que são atendidos no laboratório, na maioria com diagnóstico presuntivo de doenças autoimunes. Os analitos foram dosados por nefelometria cinética com nefelômetro Immage 800, Beckman Coulter (Califórnia, EUA). Calculou-se o intervalo de referência pelo método não paramétrico, quer dizer, estimou-se o intervalo de confiança de 95% central de cada distribuição de valores. Os limites obtidos foram: IC95% C3= 70-165 mg/dL e IC95% C4= 14-37 mg/dL. Estes resultados foram posteriormente verificados com uma série de 20 novos doadores e foram comparáveis a valores obtidos em outras séries citadas na literatura.

ABSTRACT

Malnutrition modifies the hostfis resistance to infection, altering many physiological processes, including hematopoiesis and immunological functions. In this study, we evaluated the total complement system and its C3 component factor in animals subjected to a protein malnutrition model with lipopolysaccharide (LPS) stimulation. The cellularity of blood, bone marrow and spleen, as well as the production of C3 and CH50, were evaluated. Two-month old male Swiss mice were submitted to protein malnutrition with a low-protein diet containing 4% protein. A control group received a control diet containing 20% protein. When the experimental group had reached a loss of about 20% in their original body mass, they were intravenously inoculated with LPS. Cells in the blood, bone marrow and spleen were counted and the circulating levels of C3 and CH50 were evaluated in these animals. Malnourished animals presented anemia, leucopenia, and a severe reduction in bone marrow and spleen cellularity. The survival rate of the malnourished animals was lower, as well as the production of C3 and CH50, if compared to the control animals. These findings suggest that malnourished mice present a deficient response to LPS. The decrease in the complement synthesis may be partially responsible for the immunodeficiency observed. These data lead us to conclude that the nutritional status interferes in the immune response activation.


La desnutrición proteico-energética modifica la resistencia a la infección, alterando numerosos procesos fisiológicos, incluyendo la hematopoyesis y la función inmunológica. En este estudio medimos las concentraciones séricas del factor C3 y del Sistema Complemento Total (CH50) en ratos con desnutrición proteico-energética estimulados con lipopolisacárido (LPS). Fue evaluada la celularidad periférica, medular y esplénica. Ratos Swiss, machos, adultos, con dos meses de edad fueron sometidos a desnutrición proteica con una dieta de 4% de proteína .El grupo control consumía una dieta con 20% de proteínas. Cuando los animales desnutridos perdieron 20% de su peso inicial, fueron inoculados por vía endovenosa con LPS. Fueron determinadas las concentraciones de células sanguíneas, de la médula ósea, del bazo y los valores de C3 y CH50 circulantes en los animales estimulados. Los animales desnutridos presentaron anemia y leucopenia además de una reducción significativa de celularidad de la médula ósea y del bazo. La sobrevivencia de este grupo era menor y también eran más bajas las concentraciones del factor C3 del complemento y del complemento total, en relación a los animales del grupo control. Los resultados sugieren que animales desnutridos muestran una respuesta deficiente a LPS. La síntesis menor de complemento puede ser en parte responsable por la inmunodeficiencia observada. Estos resultados nos conducen a inferir que la desnutrición proteico-energética interfiere en la activación de la respuesta inmune.

Neste estudo, avaliamos as concentrações séricas do fator C3 e do Sistema Complemento total (CH50) em um modelo no qual camundongos submetidos à desnutrição proteico-energética são estimulados com lipopolissacarídeo (LPS), e avaliamos a celularidade periférica, medular e esplênica. Camundongos Swiss, machos, adultos, com dois meses de idade foram submetidos ao processo de desnutrição proteica com uma dieta contendo 4% de proteína em comparação aos animais controles com uma dieta contendo 20% de proteína. Quando os animais do grupo desnutrido alcançaram aproximadamente 20% de perda de peso, em relação ao inicial, foram inoculados endovenosamente com LPS. As células do sangue, da medula óssea e do baço foram quantificadas, bem como as concentrações circulantes de C3 e CH50 em animais estimulados com LPS. Os animais desnutridos apresentaram anemia e leucopenia, além de redução significativa da celularidade da medula óssea e do baço. Os animais desnutridos apresentaram menor taxa de sobrevivência, bem como das concentrações do fator C3 do complemento e do complemento total em relação aos animais controles. Estes resultados sugerem que animais desnutridos apresentam uma resposta deficiente aos LPS. A síntese menor do complemento pode ser em parte responsável pela imunodeficiência observada. Estes resultados conduzem-nos a inferir que a desnutrição proteico-energética interfere na ativação da resposta imune.

ABSTRACT

The study was carried out on 27 patients with psoriasis and 9 heatlhy volunteers. Results showed no difference between IgA residues in various skin tissues of psoriasis patients. There is an increase of IgM residue in psoriasis lesions, but in healthy subjects there is not. In the lesions after the treatment and in the tissue without sporiasis, IgM residue is less than in the lesions before treatment. There is an increase of IgG residue in the lesion but in healthy skin there is not. IgG residue in healthy skin is less than in psoriasis. After the treatment, in psoriasis skin lesion, IgG is reduced slowly in comparing with that before the treatment C3 complement residue in the lesions is more than in healthy skin
